Querrey lost 6-4, 6-7 (10), 7-6 (5) to Czech Jiri Vesely, two-time champion Isner lost 7-6 (3), 5-7, 6-2 to Hyeon Chung of Korea and Johnson was beaten 2-6, 6-2, 6-1 by fifth-seeded former champion Roberto Bautista Agut of Spain.
Bautista Agut, the world No.21, will go on to face Robin Haase in Friday's semi-finals after the Dutchman's defeat of giant-killing German Peter Gojowczyk 6-4 6-4.
"I had luck to break his serves twice and that's it, then I held my serve in all the second set, saving break points, playing good tennis down the line, and I had to take a risk playing against David because he's a fighter", del Potro said.
